Zusammenfassung
Einleitung: Kurative und palliative Anwendungen von Mistel -
extrakten gehören in der komplementären onkologischen
Medizin zu den häufigsten Behandlungsverfahren. Auch für
Löwenzahnextrakte konnten bereits antineoplastische Effekte
auf Tumorzelllinien der Erwachsenenonkologie beschrieben
werden. Für Kinderkrebszelllinien liegen hingegen bislang keine
Daten vor. Des Weiteren sind therapeutische Kombinations -
effekte von Mistel- und Löwenzahnzubereitungen noch unzureichend
untersucht.
Methoden: Zytotoxische Effekte einer Taraxacum-officinale-
Zubereitung (WALA) und der Mistelpräparate Iscucin® Tiliae
und Iscucin® Pini (WALA) wurden anhand von Zellviabilitäts-
Assays an 18 pädiatrischen und 14 adulten Tumorzelllinien
untersucht. Zudem wurden die Auswirkungen auf die Zellpro -
liferation, Zellzyklusverteilung, Apoptoseinduktion, mitochondriale
Integrität sowie auf die Tumorzellmigration von aus -
erwählten Neuroblastom- und Ovarialkarzinomzelllinien
ermittelt. Außerdem wurden Kombinationseffekte einer
Behandlung aus Mistel- und Löwenzahnextrakt analysiert.
Ergebnisse: Die Viabilität aller getesteten Tumorzellen wurde
sowohl durch die Löwenzahnzubereitung als auch durch die
beiden Mistelpräparate dosisabhängig verringert. Für Neuroblastom-
und Ovarialkarzinomzellen konnte eine Apoptoseinduktion,
eine Abnahme der mitochondrialen Integrität sowie
eine Abschwächung der Tumorzellmigration beobachtet werden.
Kombinationen aus Mistel- und Löwenzahnzubereitungen
zeigten überwiegend synergistische Effekte.
Schlussfolgerung: Die Ergebnisse liefern erste präklinische
Daten für tumorzellspezifische zytotoxische und antimetasta -
tische Effekte einer wässrig-fermentierten Löwenzahnzubereitung
für ein breites Spektrum von pädiatrischen und adulten
Tumorzelllinien. Misteltherapien können durch die Gabe von
Löwenzahnextrakt unterstützt werden.
Abstract
Introduction: Applications of mistletoe are among the most
common treatment methods in complementary oncological
medicine. Antineoplastic effects on adult tumor cell lines have
also been reported for dandelion preparations. However, no data
are yet available for pediatric cancer, or combinational effects of
mistletoe and dandelion therapies.
Methods: Cell viability upon treatment with Taraxacum officinale
preparation (WALA) and mistletoe preparations Iscucin® Tiliae
and Iscucin® Pini (WALA) were investigated on 18 pediatric and
14 adult tumor cell lines. Effects on proliferation, cell cycle distribution,
apoptosis induction, mitochondrial integrity and migration
of selected neuroblastoma and ovarian cancer cell lines and
combinational effects of mistletoe and dandelion extract were
analyzed.
Results: Cell viability of all tumor cell lines was dose-dependently
reduced by the dandelion preparation as well as by the two
mistletoe preparations. For neuroblastoma and ovarian carcinoma
cells, apoptosis induction, mitochondrial integrity decrease
and attenuation of migration was observed. Combinations
of mistletoe and dandelion showed predominantly synergistic
effects.
Conclusion: The results provide first preclinical data for tumor
cell-specific cytotoxic and anti-metastatic effects of dandelion
preparations for a broad panel of pediatric and adult tumor cell
lines. Combination of preparations from mistletoe and dandelion
therapies showed supportive effects of the latter on mistletoe
applications.
